| Overview | ethyl 4-methoxyacetoacetate is a colorless transparent liquid with aromatic odor, which is an important pharmaceutical intermediate compound, at present, these compounds are mainly used for the synthesis of a new anti-AIDS drug dulutovir, which is an anti-AIDS integrase inhibitor approved by the FDA of GlaxoSmithKline in 2013, the trade name is Tivicay. Dulutvir is the 4th new drug listed by GSK company in 2013 as a drug to be evaluated by FDA, with sales expected to reach US $1.62 billion in 2020. The safety of the drug is improved compared with existing HIV integrase inhibitors retegavir and ertegavir. Ethyl 4-methoxyacetoacetate is used as the starting material for the synthesis of dulutevir, and its quality and price have a great influence on the further synthesis of dulutevir. Therefore, the study on the synthesis of ethyl 4-methoxyacetoacetate has certain significance. |
| preparation method | a synthesis method of ethyl 4-methoxyacetoacetate, the specific steps are as follows: (1) add tetrahydrofuran into the reaction kettle in advance, enter the inert gas, set the internal temperature 15-25 ℃, add sodium hydride under stirring condition, and continue to add tetrahydrofuran after adding, the mixture of methanol and ethyl 4-chloroacetoacetate was added dropwise and reacted for 4-6h under the control of system temperature of 20 ℃, and then the reaction was continued for 3-5H when the temperature was increased to 20-25 ℃; (2) the reaction system is cooled to -7-0 ℃, and hydrochloric acid solution with a molar concentration of 2mol/L is added under stirring to adjust the pH of the reaction system to 11-13, the solid in the reaction system increased significantly, and the reaction liquid was filtered to obtain a white solid; (3) the obtained white solid was added to ethyl acetate, hydrochloric acid solution with a molar concentration of 6mol/L was added dropwise at 0 ℃, the solid was gradually dissolved, the pH of the reaction system was adjusted to 2-4, the reaction solution was filtered, and the filtrate was separated from the organic layer, the organic phase was decolorized by decolorizing agent and then the solvent was distilled off to obtain pure ethyl 4-methoxyacetoacetate. |
